**Human Protein C (Protein C) ELISA Kit Instruction Manual**
This ELISA kit is designed for the quantitative determination of Human Protein C in various biological samples. It is intended for research use only and should not be used for diagnostic or therapeutic purposes.
**Kit Specifications:**
- **Configuration:** 48-well or 96-well format
- **Standard Dilution:** 1.5 mL × 1 bottle
- **Enzyme Standard Reagent:** 3 mL × 1 bottle (for 48-well) / 6 mL × 1 bottle (for 96-well)
- **Storage Conditions:** All reagents should be stored at 2–8°C, except for the chromogen B solution, which should be stored at -20°C
**Calculation Method:**
To determine the concentration of Protein C in your samples, follow these steps:
1. Plot a standard curve by plotting the concentration of the standard on the x-axis and the corresponding OD values on the y-axis.
2. Draw the standard curve on graph paper or using software.
3. Determine the OD value of your sample from the standard curve to find its corresponding concentration.
4. Multiply this value by the dilution factor if your sample was diluted before testing.
5. Alternatively, calculate the linear regression equation from the standard concentrations and their OD values. Use this equation to calculate the sample concentration based on its OD value, then multiply by the dilution factor to obtain the actual sample concentration.
**Kit Components:**
- **Sealing Film:** 2 pieces (for 48-well) / 2 pieces (for 96-well)
- **Standard:** 2700 ng/L, 0.5 mL × 1 bottle
- **Enzyme Standard:** 1×48 / 1×96
- **Sample Diluent:** 3 mL × 1 bottle (for 48-well) / 6 mL × 1 bottle (for 96-well)
- **Developer A:** 3 mL × 1 bottle (for 48-well) / 6 mL × 1 bottle (for 96-well)
- **Chromogen B Solution:** 3 mL × 1 bottle (for 48-well) / 6 mL × 1 bottle (for 96-well), store at -20°C
- **Stop Solution:** 3 mL × 1 bottle (for 48-well) / 6 mL × 1 bottle (for 96-well)
- **Concentrated Washing Solution:** 20 mL × 20 times (for 48-well) / 20 mL × 30 times (for 96-well)
**Storage Instructions:**
All components should be stored at 2–8°C unless otherwise specified. The chromogen B solution must be kept at -20°C to maintain stability and performance.
**Preparation and Usage:**
Before starting the assay, ensure all reagents are brought to room temperature. Carefully follow the step-by-step protocol provided in the manual. Proper handling and accurate pipetting are essential for reliable results.
**Note:**
This kit is intended for in vitro diagnostic research use only. Do not use it for human or animal clinical diagnosis. Always wear appropriate personal protective equipment when handling reagents. Dispose of waste according to local regulations.
